I love this scene so much. By saying “You did the best you could, I forgive you”, Anna is showing Dean the kind of mercy heaven never showed her. And that look on Dean’s face, okay, jfc. He just came back from 40 years in hell where he did unspeakable things, and this fallen angel right here is the first person to tell him this thing he needs to hear with the sincerity he doesn’t think he deserves. For all this talk of heaven’s mercy it ends up coming from heaven’s fugitive, from Anna’s aspirations of humanity. She wanted to be compassionate and merciful, to be different from other angels, as much as Dean needed to hear that compassion and mercy. Don’t they both want to be better than the sins they’ve been accused of?

I think there’s just a whole world of layered meanings in this little scene.  It’s meant for Dean and Anna for all the reasons you just said, and it would have stood as a great scene for that reason alone if it had been, say, just Uriel, or Uriel and Random Garrison-Mate #12 there to kill Anna.

But because it’s Cas there, it’s also meant to demonstrate to him why Anna chose to become human and how their way is better—and, as becomes clear a moment later when he offers his own rather flat attempt at an apology, to make the point that unlike Dean, he is not forgiven.  He’s moved enough by what he sees to know that he wants for himself what Anna has just given Dean, but doesn’t yet understand why he does, or why he should, or what he needs to do to earn it (i.e., to really, truly repent what he’s about to do; not just to wish he didn’t have to, but to acknowledge that he’s wronging her.)

To me that’s what that little downward glance was always about.  Not jealousy that Anna was kissing “his” human, but an unexpected pang of guilt or regret and a desire to be absolved.  I think that’s the whole reason Cas did try to apologize right at that moment.  It must have confused him no end when he said exactly the same thing that Dean did (and you can hear him hesitating just a little, trying the unfamiliar words on for size; ‘How do apologize?’) but got a completely different response.
